Chitin and its derivatives in recent time gained industrial and bio medical applications which created growing demand for chitin extraction from new sources. The present study is aimed at extracting chitin from fish scale and develop antibiotic-loaded chitin film against diabetic foot ulcer causing bacteria. Chitin was extracted from scales of four fish species using chemical methods and yield of 2.240 ± 0.04, 3.260 ± 0.94, 2.930± 0.47 and 5.230±0.81 g/10g reported from Lates calcarifer, Sardinells longiceps, Labeo rohita and Oreochromis niloticus, respectively. Structural characteristics of the chitin is provided by FTIR data analysis. The SEM images of chitin exhibited smooth and thick surface morphology. Antimicrobial activity of chitin film against diabetic foot ulcer causing pathogens is tested. Chitin from S. longiceps +Ambistryn showed high inhibition of 3.1, 3.5, 3.1 and 3.0 cm in the clinical pathogens, Escherichia coli, Klebsiella pneumoniae, Candida albicans, Staphylococcus aureus and Enterococcus faecalis, respectively. Chitin extracted from fish scale is an effective antibacterial agent which inhibits growth of clinical pathogen. The S. longiceps chitin + Ambistryn film produced higher antibacterial activity than other chitin film. In vitro studies suggested potential antibacterial mechanisms of chitin and it’s nontoxic nature. Chitin from fish scales could be considered in drug formulation against bacterial infection in diabetic foot ulcer treatment.
